Non-genomic effects of thyroid hormones on mesenchymal stem cells in tumour angiogenesis

Schmohl Kathrin A , Dohmann Maike , Schwenk Nathalie , Bartenstein Peter , Nelson Peter J , Spitzweg Christine

Tumour stroma formation is associated with angiogenesis and requires interactions of various different cell types, including endothelial cells and mesenchymal stem cells (MSCs), which are actively recruited into growing tumour stroma. Thyroid hormones T3 and T4 act as non-classical proangiogenic modulators mediated by non-genomic mechanisms via cell surface receptor integrin αvβ3. The deaminated T4 derivative tetrac is a specific inhibitor of thyroid hormone action a...

Non-genomic effects of thyroid hormones on endothelial cell tube formation

Schmohl Kathrin A , Dohmann Maike , Wechselberger Alexandra , Nelson Peter J , Spitzweg Christine

Wound healing and tumour stroma formation are associated with angiogenesis and require interactions of various different cell types. We and others have shown that mesenchymal stem cells (MSCs) differentiate into fibroblast-/pericyte-like cells and secrete proangiogenic factors. Thyroid hormones act as proangiogenic modulators mediated by non-genomic mechanisms via cell surface receptor integrin αvβ3. The aim of this study is to evaluate the stimulatory activity of T3...

The tumor-homing property of mesenchymal stem cells (MSC) has led to their use as delivery vehicles for therapeutic genes such as the sodium iodide symporter (NIS), which has convincingly been demonstrated by our recent studies of systemic NIS gene delivery using MSCs as delivery vehicles in both subcutaneous and orthotopic xenograft mouse models. External beam radiation therapy (EBRT) represents a promising tool in the application of engineered MSC (eMSCs)-based gene therapy ...

Background: Treatment resistance in prostate cancer can be the result of multiple different tumour cell adaptations generating castration-resistant prostate cancer (CRPC). Despite advancements in treatment, CRPC remains a highly lethal disease. Some CRPCs become AR independent with loss of AR expression and lineage plasticity, urgently requiring novel therapeutic strategies. BCL2 can be upregulated in some CRPCs and may be a therapeutic target for this disease subset.<p cl...
